The grandparent DNA test starts by collecting buccal (mouth) swab samples from the grandparent(s) and grandchild. These swabs gather cells containing each individual’s unique DNA. In the lab, DNA is extracted from the cells and specific regions are amplified via PCR (polymerase chain reaction) for analysis. The grandparent’s DNA pattern is then compared to the grandchild’s pattern.
A statistical analysis is performed to calculate the likelihood of a biological grandparent-grandchild relationship based on the genetic markers shared between them. This analysis produces a “grandparentage index” – the higher the index value above 1.0, the more likely the grandparent and grandchild are biologically related. An index below 1.0 indicates a lower probability of a biological relationship.
